چکیده

We describe a new cation-exchange fluorometry procedure for separation and measurement of three 5-hydroxyindole metabolites of tryptophan in blood. The metabolites are detected and measured via the fluorescence emitted in the visible region when the separated metabolite is added to a hydrochloric acid-ascorbic acid mixture. The assay requires only 30 mm and is both selective and sensitive. The normal concentration of 5-hydroxytryptamine (serotonin) in serum is 98 ag/liter; 5-hydroxyindole acetic acid and 5-hydroxytryptophan are normally present in concentrations of less than 20 pg/liter.

Tryptophan metabolism via serotonin in rats with hexachlorobenzene experimental porphyria.

One of the three pathways for the metabolisation of dietary tryptophan is the formation of serotonin. Tryptophan hydroxylase catalyses the formation of 5-hydroxytryptophan, the first and regulatory step of this biosynthesis. The aim of the present work is to study alterations in this tryptophan metabolism in rats with experimental Porphyria Cutanea Tarda induced by hexachlorobenzene. With this ...

Fate of Dietary Tryptophan in Young Japanese Women

The purpose of this study was to determine, using the high-performance liquid chromatographic methods recently modified by us, the fate of dietary tryptophan in 17 healthy female Japanese adults who ate self-selected food. The experimental period was 22 days. The habitual intake of tryptophan was 3328.4 μmol/day. 24-hour urine samples were collected at the beginning of the experiment and then o...

Dissimilation of tryptophan and related indolic compounds by ruminal microorganisms in vitro.

Intraruminal doses of L-tryptophan cause acute pulmonary edema and emphysema in cattle. The D and L isomers of tryptophan and 22 related indolic compounds were incubated with ruminal microorganisms in vitro. Incubation of L-[U-benzene ring-(14)C]tryptophan with ruminal microorganisms for 24 h resulted in 39% of the added radioactivity being incorporated into skatole, 7% into indole, and 4% into...

Melatonin formation in pineal gland from rats with hexachlorobenzene experimental porphyria.

Hexachlorobenzene produces an experimental hepatic porphyria in rats, which is similar to human porphyria cutanea tarda, with hyperpigmentation as one of its characteristic features. Alterations in tryptophan metabolism have been previously observed in this chronic porphyria. Melatonin formation from tryptophan via serotonin shows diurnal rhythmicity in the pineal gland, and higher values are o...

The Biosynthetic Pathway of Indole-3-Carbaldehyde and Indole-3-Carboxylic Acid Derivatives in Arabidopsis.

Indolic secondary metabolites play an important role in pathogen defense in cruciferous plants. In Arabidopsis (Arabidopsis thaliana), in addition to the characteristic phytoalexin camalexin, derivatives of indole-3-carbaldehyde (ICHO) and indole-3-carboxylic acid (ICOOH) are synthesized from tryptophan via the intermediates indole-3-acetaldoxime and indole-3-acetonitrile. Based on feeding expe...
